Yeast ADH1 +sqr composite promoter
We understand that the sqr promoter BBa_K4743019,originally from prokaryotes and sensitive to hydrogen sulfide, cannot function in yeast due to their eukaryotic nature. To address this, we fused the ADH1 promoter, which drives RNA polymerase activity in yeast, with the sqr promoter. In this arrangement, ADH1 acts as the driver, while sqr serves a regulatory function by aiding in the binding of the sqrR repressor. Nevertheless, the functionality of this composite promoter requires verification.
